Ricin toxin binding subunit B (RTB) is one of the subunits of the ricin protein. RTB has been used as adjuvant, but little is known about its mechanism. In this study, we found that RTB increased not only nitric oxide (NO) release, but also tumor necrosis factor (TNF)-α and interleukin (IL)-6 production in mouse macrophage cell line RAW264.7 cells. They subsequently exhibited enhanced ConA-induced T-cell and LPS-induced B-cell proliferative responses. We also examined the cytokines that were produced from splenocytes following in vitro RTB administration. Increased levels of IL-2, interferon (IFN)-γ and TNF-α were observed, while IL-4 and IL-5 were unaffected. These results demonstrate that recombinant RTB can act on the immune system and activate T-cells by introducing a Th1 immune response. Th1 cells might be the primary cellular target affected by RTB. Our results suggest that the recombinant RTB can promote the activation of macrophages and has a beneficial effect on immunomodulatory activity. 相似文献

A full-length cDNA coding for hydroperoxide lyase (CsHPL) was isolated from cucumber fruits of No. 26 (Southern China type) and No.14-1 (Northern China type), which differed significantly in fruit flavor. The deduced amino acid sequences of CsHPL from both lines show the same and significant similarity to known plant HPLs and contain typical conserved domains of HPLs. The recombinant CsHPL was confirmed to have 9/13-HPL enzymatic activity. Gene expression levels of CsHPL were measured in different organs, especially in fruits of different development stages of both lines. The HPL activities of fruit were identified basing on the catalytic action of crude enzyme extracts incubating with 13-HPOD (13-hydroperoxy-(9Z,12E)-octadecadienoic acid) and 13-HPOD + 9-HPOD (9-hydroperoxy-(10E,12Z)-octadecadienoic acid), and volatile reaction products were analyzed by GC-MS (gas chromatography-mass spectrometry). CsHPL gene expression in No. 26 fruit occurred earlier than that of total HPL enzyme activity and 13-HPL enzyme activity, and that in No. 14-1 fruit was consistent with total HPL enzyme activity and 9-HPL enzyme activity. 13-HPL enzyme activities decreased significantly and the 9-HPL enzyme activities increased significantly with fruit ripening in both lines, which accounted for the higher content of C6 aldehydes at 0–6 day post-anthesis (dpa) and higher content of C9 aldehydes at 9–12 dpa. 相似文献

Optical and photoelectrochemical (PEC) properties of a TiO2 thin film electrode doped with a new variation of ruthenium–(4,4′dimethyl-2,2′-bipyridine)–isothiocyanato–tungsten[bis-(phenyl-1,2-ethilenodithiolenic)] bimetallic complex (BM) were investigated. Physical adsorption process was used to immobilise the BM on the TiO2 thin film. Crystalline structure and surface morphology of the thin films were examined using scanning electron microscopy (SEM), X-ray diffraction (XRD) and energy-dispersive X-ray (EDX) techniques. N3 commercial dye was also used as a dopant to the TiO2 films for comparison. Light absorption spectra and bandgap energy of the thin films were determined using UV–vis spectroscopy. Light absorption of the TiO2 thin film doped with BM was better than the TiO2 doped with the N3 commercial dye. Band edges of the TiO2 thin film and the BM were determined via cyclic voltammetry (CV) measurements. Top-edge of the BM valence band (VB) was more positive than the bottom edge of the conduction band (CB) of the TiO2 film (vs. NHE). PEC analysis indicated that photocurrent of TiO2 doped with the BM electrode was higher than TiO2 doped with the N3 in the beginning of illumination process, but the performance was defeated after a while. Based on the optical properties and the PEC analyses, BM has potential to be used as dye sensitisers for a PEC cell. 相似文献

The ultraviolet radiation aging behaviors of PVC/CaCO3 and PVC/CaCO3/macromolecular modifier composites were studied through whiteness measurement, Fourier transform infrared spectroscopy, ultraviolet–visible spectroscopy, scanning electron microscopy, and mechanical properties test. It was found that nano‐CaCO3 particles used as ultraviolet light screening agents could significantly enhance the antiaging properties of PVC materials. Due to the macromolecular modifier coated on nano‐CaCO3 particles, the compatibility of nano‐CaCO3 and PVC matrix was improved, resulting in uniform dispersion of nano‐CaCO3 in PVC matrix. Therefore, the PVC/CaCO3/MP composite exhibited better antiaging properties than PVC/CaCO3 composite. After 12 h of ultraviolet irradiation, the tensile strength retention, elongation at break retention, and impact strength retention of PVC/CaCO3/MP composite were 79.5%, 74.5%, and 75.3%, which were much higher than that of neat PVC and PVC/CaCO3 composite. © 2012 Wiley Periodicals, Inc. Amphiphilic heteroarm star‐shaped polymers have important theoretical and practical significance. In this work, amphiphilic heteroarm star‐shaped polymer was synthesized by the use of polyfunctional chain transfer agent via sequential free radical polymerization in two steps. First, conventional free radical polymerization of methyl methacrylate (MMA) initiated by 2,2′‐azobis (isobutyronitrile) (AIBN) was carried out in the presence of polyfunctional chain transfer agent, pentaerythritol‐tertrakis (3‐mercaptopropinate) (PETMP). At appropriate monomer conversion, about two‐arm s‐PMMA having two residual thiol groups at the chain center was obtained. Second, the s‐PMMA obtained above was used as macro‐chain‐transfer agent for free radical polymerization of acrylic acid (AA). The heteroarm star‐shaped polymer with the hydrophobic PMMA segment and the hydrophilic PAA segment was obtained. The successful synthesis of heteroarm star‐shaped polymers, (PMMA)2(AA)2, was confirmed by 1H‐NMR and its self‐assembly behavior in different solvents. © 2012 Wiley Periodicals, Inc. J. Appl. Polym. Crosslinkable polytriazole resins (CPTs) were synthesized by 1,3-dipolar cycloaddition reaction between azide and alkyne monomers. Adhesion properties of CPTs and an epoxy resin to different metal substrates (copper, iron and aluminium) were evaluated in terms of the tensile lap shear strength. CPTs to aluminium and iron substrates have higher adhesion strength than those to copper substrates. The effect of temperature and humidity on adhesion strength has been investigated. The resin CPT-3 has the best heat resistance among CPTs, whose retentions of adhesion strength are approximate 80% at 150?°C and 70% at 180?°C to all the substrates. CPTs have high adhesion properties at high temperature and humidity as compared with an epoxy resin. 相似文献

One of the important characteristics of the thermosetting resins such as urea-formaldehyde resin is the cure property. This is an invaluable characteristic for determining the processing capabilities of the resin. Differential scanning calorimetry (DSC) measurements are ideally suited to achieve this end. This paper presents the results of the effect of process variables on the enthalpy of cure of a low formaldehyde emission UF resin prepared by a four-stage process as described in the text. Response surface methodology (RSM) was used to design and the analyze the experiments. Urea (U) was added to the reaction mixture according to a pre-determined program of varying the number of additions and the duration between the additions during the second stage of the process. A mathematical relationship between the process variables and enthalpy of cure was established. The urea-formaldehyde (UF) resin was prepared under optimum conditions. Experiments were performed with the resin so prepared to determine the energy of activation of the curing reaction catalyzed by ammonium chloride. 相似文献
